Before Jefferson City

The Secretary of State’s bicentennial page says St. Charles was the temporary seat of government from 1821 until 1826, in a building that is now a state historic site.

What We Could Not Verify

Why the seat was temporary beyond Jefferson City being planned as the permanent capital, and the exact move date, are not stated on that page. The history timeline answer gives October 1, 1826.
